Development of Loop-Mediated Isothermal Amplification Assay for Rapid Detection and Analysis of the Root-Knot Nematode Meloidogyne hapla in Soil

Soil analysis is crucial for estimating the risk of crop damage by root-knot nematode Meloidogyne hapla. Here, we developed an assay based on Loop-mediated Isothermal Amplification (LAMP). The LAMP primers were verified specificity against 10 different species. A manual soil DNA extraction, referred to as SKMM, was and compared with a FastDNA kit followed purification. extracted both methods from artificially inoculated soils well naturally infested collected farm fields. exclusively amplified M. hapla colorimetric real-time LAMP. detection limit 193 gene copies 0.0016 juveniles (12 pg µL−1) per reaction. concentrations purity (A260/A230) significantly higher using SKMM procedure kit.
